This game marks the return of Alex and Urien, both of whom last appeared canonically in 3rd Strike. Both characters were also featured in relatively obscure spin-off games; Alex was in Tatsunoko VS Capcom, and Urien was in Street Fighter X Mega Man.
R. Mika's tag team partner is Yamato Nadeshiko, another wrestler who works for the same wrestling organization. Her appearance and background date back to a volume of the Capcom Secret File released in 1998.

Going by Mel's canon age and appearance, the story takes place at least a year after Super Street Fighter IV, but before Street Fighter III: New Generation.
Necalli was originally meant to eat his opponent's bodies after winning but Capcom felt that this was too mature so they made Necalli eat their souls instead.
A character based on Momotaru from Capcom's puzzle game Pirate Ship Higemaru was considered as a playable character for the game. Birdie was chosen over him in the end, however. Momotaru was instead given a profile in the Street Fighter V character guide in August 2016.
The game originally had a more realistic look during it's early stages of development.
The idea of a fighter who uses poison in the Street Fighter games was originally considered during the development of Street Fighter III: 3rd Strike, when the team members had a debate on including Remy (a character with a playstyle similar to Guile's) or a different, poison-using character.
Capcom eventually decided to implement this idea with the SFV newcomer, F.A.N.G.
Aside from Pullum Purna in the non-canon Street Fighter EX games, Rashid is the only Middle Eastern character in the series story.

Chun Li's Critical Art does 37 hits. This is a shout-out to Evo Moment #37, the famous YouTube 3rd Strike video taken at Evolution 2004, in which Daigo Umehara perfectly parried Justin Wong's Chun-Li Super Art.
When set on Color #2, Karin's alternate costume is a shout-out to Roll from the Mega Man series.
Laura is the older sister of Sean, a playable character and Ken's student in Street Fighter III.
In story mode, Karin receives information about an old hermit and Senjutsu master who fights with his dominant hand sealed. This is a direct reference to Oro, who last appeared in 3rd Strike. He is canonically one the strongest fighters in the series and a future mentor for Ryu.

R. Mika's V-Reversal is visually identical to the Stone Cold Stunner, which was Steve Austin's iconic finishing move.
The wrestling magazine Zangief is reading in story mode has the same cover as the Street Fighter Alpha 3 volume of the Capcom Secret File, which was released in 1998.
In his prologue story mode, one of the text messages on Rashid's social media feed is from H.W.A._Epic. This is a reference to Hugo's endings in both Ultra Street Fighter IV and 3rd Strike, in which Hugo and Poison team up and start the Huge Wrestling Army.
